@article{oai:teapot.lib.ocha.ac.jp:00035010, author = {山崎, 順啓 and Yamazaki, Nobuhiro}, issue = {1}, journal = {お茶の水女子大學自然科學報告}, month = {Jul}, note = {application/pdf, 紀要論文, Feulgen,辻の法により抽出したDNAおよびそれにpancreatinを作用させて得たb核酸につき,沈降,拡散,光散乱,粘度の測定を行い,両者を物理化学的性格を中心にして種々な角度より比較検討を行った。その結果1.沈降実験よりDNA S_0:28.8×10^<-13>(cm/sec)/(dyne/g) b核酸S_0:3.5×10^<-13>(cm/sec)/(dyne/g) 2.拡散実験よりDNA D_0:0.2×10^<-7>cm^2/sec b核酸 D_0:3.25×10^<-7>cm^2/sec 3.両定数よりの分子量DNA:700×10^4 b核酸:6×10^4 4. 光散乱実験よりDNA分子量:200〜1000×10^4 DNA分子の大きさ:3000Å (End to end distance) b核酸分子量:4〜10×10^4 b核酸分子の大さ:600Å (End to end distance) 5. 粘度実験よりDNA固有粘度:2〜3 b核酸固有粘度:0.26の値を得た。以上の結果および沈降図形,拡散曲線,光散乱強度の角度分布の解析より次のことが考察された。すなわちFeulgen DNAは球形に近い形のやや不均一な,しかも濃度依存性の高い分子で粒子間の相互作用は大きい。b核酸は比較的均一な濃度依存性の低い粒子間の自由度が高い解重合体である。以上でDNAがb核酸に変化した場合の物理化学的相違は明にされたがDNAのどの結合がPancreatinの中の如何なる酵素により切離されるかを解明することが今後著者等に残された課題であろう。, As is already known high molecular D・N・A is digested by ordinary pancreatin with a gradual fall of the viscosity which stops after 26 hours. The degree of solubility in the acid of low molecular D・N・A(b-nucleic acid) thus obtained is about the same as the parent D・N・A. As a result of my experiment by ultraviolet absorption spectra, migration rate of paper electrophoresis and measurement of acidity, the b-nucleic acid seems to be a kind of depolymerized substance of D・N・A which is cut off by the pancreatin at certain points of the polymer chains. For the purpose of finding out the true nature of the b-nucleic acid, the difference between D・N・A and b-nucleic was further investigated by experiments with chemicophysical properties such as sedimentation, diffusion, lightscattering, and viscosity. The results of these experiments are as follows : 1) Sedimentation constant (with Spinco Model E ultracentrifuge) parent D・N・A : So=28.8×10^<-13> (cm/sec)/(dyne/g) b-nucleic acid : So=3.5×10^<-13> (cm/sec)/(dyne/g) 2) Diffusion constant (with Neurath type cell) parent D・N・A : D_0=0.2×10^<-7> cm^2/sec b-nucleic acid : D_0=3.3×10\ ^<-7> cm^2/sec 3) Light scattering parent D・N・A : M=200〜1000×10^4 R=3000Å b-nucleic acid : M=4〜10×10^4 R=600Å 4) Viscosity (with ubbelohode viscometer) parent D・N・A : [η]=2〜3 b-nucleic acid : [η]=0.3 5) Molecular weight calculated from Sedimentation constant and Diffusion constant parent D・N・A : M=700×10^4 b-nucleic acid : M=6×10^4 As a result of the above experiments it was found out that the b-nucleic acid is a lower molecular Dノチ which was separated into small pieces of about 1/100 of the original D・N・A, having a more uniform, symmetrical shape than the original D・N・A. It was also found out that when these small pieces of b-nucleic acid were put into a saline solution, they were not affected by each other as the parent D・N・A. The next problem is to find out the pure enzime in the pancreatin which cuts the D・N・A chain at certain points and to find out where it cuts the chain.}, pages = {86--125}, title = {b核酸に関する物理化学的研究}, volume = {11}, year = {1960} }
