B処理卵ではGVは移動しなかったが、nocodazole処理卵ではGVが卵表から離れて、求心方向へと移動した。\\\\r\\\\\\<br/>　蛍光抗体染色により微小管構造を観察すると、予定動物極によく染色される点状構造とそこから放射状に広がる構造、細胞質の網目状の構造と、GVの周囲を取り囲む構造が観察された。nocodazole処理卵ではこれらの構造はいずれも観察されなかった。\\\\r\\\\\\<br/>　共焦点レーザー顕微鏡によって、三次元的に微小管構造を観察した。放射状微小管は動物極域の中心に近いところではGVの膜に沿って存在し、途中からGVの膜を離れて表層に沿って伸び、その先端はいくつかに分岐して、細胞表面に存在する網目状構造に入り込んでいることがわかった。この微小管構造がGVを卵表に保持していることが示唆された。\\\\r\\\\\\<br/>\\\\r\\\\\\<br/>　以上のことから、卵母細胞の予定動物極へのGVの移動と付着のどちらにも、細胞骨格のひとつ、微小管が重要な役割を担っていることが明らかになった。","subitem_description_type":"Abstract"},{"subitem_description":"動物の第一次卵母細胞は第一減数分裂前期で停止している。この時期の卵母細胞の巨大な核、卵核胞（germinal vesicle; GV）は、ホルモンなどの刺激によって成熟分裂が再開すると崩壊し、その内容物が細胞質中に拡散する。成熟分裂を再開する準備が整っている第一次卵母細胞内でのGVの位置は動物の種によって決まっており、卵母細胞の中央にある場合と卵表の特定の部域に近接している場合とがある。成熟分裂が再開すると、前者ではGVが卵表へと移動し、卵表直下に到達した後に崩壊する。後者ではその場でGVの崩壊が起こる。いずれの場合も、崩壊が起るときには、GVは卵表の特定の部域に近接していることになる。このGVの卵表への接近は、成熟分裂のための小さな分裂装置を卵表直下に形成するために必須な出来事である。また、胚の発生の基本軸となる動植物軸の決定にも必要なことである。本研究では、(1)GVの卵表への移動と、(2)移動した後、卵表直下に保持される機構について調べた。\\\\r\\\\\\<br/>　\\\\r\\\\\\<br/>  (1)GVが中央にあるテツイロナマコの卵母細胞は、予定動物極に顕著な細胞質の突起（動物極突起）がある。成熟分裂が再開するとGVは動物極突起の直下まで移動し、崩壊する。微分干渉顕微鏡を用いて成熟過程初期の卵母細胞を詳細に観察すると、GVが移動するとき、動物極突起とGVとの間に繊維状の構造が形成されることがわかった。微小管の脱重合を促進するnocodazoleで処理した卵母細胞では、GVの移動は起こらず、GVは卵母細胞の中央で崩壊した。また、固いゼリー層を除去した卵母細胞では、GVが移動するときには予定動物極の卵表にくぼみができたが、nocodazole処理によりGVが移動しなかったときには、そのようなくぼみは観察されなかった。以上の結果から、GVの移動には微小管が関与し、その構造は動物極突起付近に結合していることが示唆された。\\\\r\\\\\\<br/>
